We are a commercial-stage company focused on discovering and developing medications to treat severe endocrinologic, oncologic, metabolic, and neurologic disorders by adjusting the effects of the hormone cortisol. Cortisol is crucial for the body's stress response, metabolism, immune system, and emotional stability, with levels naturally peaking upon awakening and decreasing throughout the day. We address both insufficient cortisol activity, which can cause dehydration, hypotension, shock, fatigue, and hypoglycemia, and excessive cortisol activity, known as hypercortisolism. Hypercortisolism can lead to hypertension, diabetes, impaired glucose tolerance, obesity, fatty liver disease, depressed mood, psychosis, wasting of limbs, edema, fatigue, and insomnia. Cortisol also impacts cancer by reducing immune response to oncogenesis, protecting certain cancer cells from chemotherapy, and promoting the growth of others. Pre-clinical and clinical data suggest that modulating cortisol activity may improve outcomes for patients with fatty liver disease and metabolic dysfunction-associated steatohepatitis ("MASH"), which can lead to liver fibrosis and cirrhosis. Data also indicate potential treatments for patients with amyotrophic lateral sclerosis ("ALS") through cortisol modulation. Since 2012, we have marketed Korlym in the United States for treating hypercortisolism (Cushing's syndrome). In June 2024, we launched an authorized generic version of Korlym for the same condition. Korlym's active ingredient, mifepristone, works by competing with cortisol for binding to the glucocorticoid receptor ("GR"), effectively modulating the effects of abnormal cortisol levels and release patterns without disrupting healthy cortisol functions or rhythms.
