TYRA Biosciences is a clinical-stage biotechnology company developing precision medicines for targeted oncology and genetically defined conditions, utilizing Fibroblast Growth Factor Receptor (FGFR) biology. The company employs its in-house SNÅP precision medicine platform for drug design, aiming to identify product candidates with high potency, selectivity, and tolerability. This approach has resulted in a wholly-owned pipeline of oral small molecule product candidates targeting previously difficult-to-drug areas. TYRA Biosciences' FGFR3 Programs include oral dabogratinib (formerly TYRA-300) for Urothelial Cancers and Skeletal Dysplasia. Alterations in the FGFR3 protein receptor are a validated driver in urothelial cancers, where uncontrolled activation stimulates cellular proliferation, and in skeletal dysplasia conditions, where increased FGFR3 activity in growth plate chondrocytes limits long bone growth. Oral dabogratinib was designed to be more selective for FGFR3 over FGFR1, FGFR2, and FGFR4 to reduce off-target side effects. To date, oral dabogratinib has been administered to over 100 participants across multiple clinical studies. Initial clinical proof-of-concept results were demonstrated in the SURF301 study, a Phase 1 proof-of-concept study in metastatic urothelial carcinoma (mUC). In this study, oral dabogratinib showed encouraging anti-tumor activity and was generally well-tolerated, with infrequent FGFR2- and FGFR1-associated toxicities. Data from this study informed dose selection for future Phase 2 trials.
