We are a clinical-stage biotechnology company developing a diverse range of cell therapy and small molecule product candidates. In February 2026, we expanded our pipeline by acquiring rights to a portfolio of dual-targeting chimeric antigen receptor T-cells ("CAR-T") product candidates. These CAR-T candidates are designed to potentially treat certain blood cancers, solid tumors, and immunology indications. This portfolio includes TPST-2003, an autologous CD19/B-cell maturation antigen ("BCMA") CAR-T therapy, which is currently in clinical development for relapsed or refractory multiple myeloma ("rrMM"). Our portfolio also features two clinical-stage small molecule product candidates aimed at treating certain cancer indications. One of these, amezalpat (formerly TPST-1120), has completed a Phase 2 study for first-line hepatocellular carcinoma ("HCC"). Amezalpat is ready for Phase 3 development in HCC, and we intend to pursue business development discussions to advance its pivotal development. Our second small-molecule product candidate is TPST-1495. We plan to initiate a Phase 2 study for TPST-1495 in familial adenomatous polyposis ("FAP"), with the first patient enrollment expected in 2026. The National Cancer Institute ("NCI") is expected to fund this study, which will be conducted through the Cancer Prevention Clinical Trials Network ("CP-CTNet"), allowing advancement with limited internal capital deployment. On November 19, 2025, we executed an Asset Purchase Agreement with Erigen LLC and Factor Bioscience Inc. to acquire worldwide rights, excluding China, Russia, India, and Turkey, to assets primarily related to the autologous BCMA/CD19 dual-targeting CAR T-cell therapy known as TPST-2003. TPST-2003 is currently being evaluated in a Phase 1/2a clinical study in patients with rrMM, a Phase 1/2 investigator-initiated trial ("IIT") in patients with rrMM, and a Phase 1 clinical study.
