PolyPid Ltd. is an innovative biopharmaceutical company focused on developing long-acting, controlled-release medicines that deliver therapy directly at the site of care to improve patient outcomes. The company's pipeline includes products for surgical care and metabolic diseases. Its lead product candidate is D-PLEX 100, which combines PolyPid's proprietary PLEX technology with doxycycline, an FDA-approved antibiotic. D-PLEX 100 is designed for direct administration into the surgical site during surgery, providing a prolonged and continuous release of high local concentrations of doxycycline for 30 days to prevent surgical site infections (SSIs). In June 2025, PolyPid Ltd. announced positive topline results from its pivotal SHIELD II Phase 3 study of D-PLEX 100. This study evaluated D-PLEX 100 for the prevention of SSIs in patients undergoing abdominal colorectal surgery with large incisions. D-PLEX 100 successfully met the primary and all key secondary endpoints of the study. SSIs occur in approximately 2% to 4% of all patients undergoing inpatient surgery and account for 20% of all healthcare-associated infections (HAIs) in the United States. The CDC estimates that SSIs result in up to $10 billion of additional hospital costs per year in the United States and are the costliest HAI type of event. The WHO and CDC have identified SSIs as a high priority unmet medical need.
