We are a precision oncology company focused on discovering and developing small molecule, tumor-agnostic therapies that target p53. Our approach leverages over four decades of research into p53, a tumor suppressor protein, to create a platform that generates selective, small molecule therapies. These therapies are designed to structurally correct specific mutant p53 proteins, which are found in approximately half of all cancers, to restore their normal function. Our lead product candidate is rezatapopt, an orally available small molecule. Rezatapopt is designed to potently and selectively correct p53 misfolding caused by the p53 Y220C mutation, while not affecting normal p53. The p53 Y220C mutation is associated with approximately 1% of all cancers, including endometrial, breast, non-small cell lung cancer (NSCLC), colorectal, pancreatic, and approximately 3% of ovarian cancers. Rezatapopt aims to restore the normal p53 structure by occupying a specific pocket created by this mutation. We are pursuing a tumor-agnostic development strategy for rezatapopt. We initiated a Phase 1/2 clinical trial, PYNNACLE, in October 2020. Our strategy is to seek approval through an accelerated pathway, and we believe the Phase 2 portion of the PYNNACLE trial could serve as a pivotal study. In October 2020, the U.S. Food and Drug Administration (FDA) granted Fast Track designation to rezatapopt for treating patients with locally advanced or metastatic solid tumors that have a p53 Y220C mutation.
