We are a clinical-stage biopharmaceutical company focused on developing distinct therapies for neurological, inflammatory, and immunological (NI&I) conditions with significant unmet needs. We aim to modify biological pathways linked to specific clinical impairments to alter the course of disease. Our focus is on creating selective compounds for challenging molecular pathways, and we have built a portfolio of small molecule drug candidates. Our two clinical-stage drug candidates, PIPE-791 and PIPE-307, were discovered internally. We are developing PIPE-307 in collaboration with J&J. Our wholly-owned lead asset, PIPE-791, is a novel, brain-penetrant, small molecule inhibitor of the lysophosphatidic acid 1 receptor (LPA1R). It is currently in development for idiopathic pulmonary fibrosis (IPF) and chronic pain. LPA1R antagonism is a clinically validated mechanism in IPF. In September 2025, we reported positive top-line data from our completed Phase 1b PET trial, which measured the relationship of pharmacokinetics to receptor occupancy by PET imaging. This data affirmed the planned dose selection for our Phase 2 trial of PIPE-791 in IPF, which began in December 2025. In the fourth quarter of 2025, we completed enrollment for a Phase 1b study exploring the safety and efficacy of oral PIPE-791 in subjects with chronic osteoarthritic pain or chronic low back pain. We anticipate top-line data from this trial in the second quarter of 2026. Our second drug candidate, PIPE-307, is a selective, small-molecule inhibitor of the muscarinic type 1 receptor (M1R). It is in development for depression and relapse-remitting multiple sclerosis. We have completed two Phase 1 trials of PIPE-307 in healthy volunteers.
