Ventyx Biosciences is a clinical-stage biopharmaceutical company focused on developing new oral treatments for autoimmune, inflammatory, and neurodegenerative diseases. The company uses its expertise in medicinal chemistry, structural biology, and immunology to discover small molecule drugs for conditions with significant unmet medical needs. Ventyx Biosciences is developing a portfolio of small molecule inhibitors targeting the NOD-like receptor protein 3 (NLRP3) inflammasome. This includes VTX2735, a peripherally restricted NLRP3 inhibitor, and VTX3232, a CNS-penetrant NLRP3 inhibitor. In the first quarter of 2024, Ventyx Biosciences reported positive topline results from a Phase 2 proof-of-concept trial of VTX2735 in cryopyrin-associated periodic syndromes (CAPS) patients. The company plans to develop VTX2735 further for cardiovascular diseases, initially focusing on recurrent pericarditis, and initiated a Phase 2 trial for this indication in January 2025. Topline results are expected in the second half of 2025. For VTX3232, positive topline results from a Phase 1 trial in adult healthy volunteers were reported in the first quarter of 2024. In August 2024, a Phase 2 trial of VTX3232 was initiated in participants with early Parkinson's disease, with topline results anticipated in the first half of 2025. Additionally, in December 2024, a Phase 2 trial of VTX3232 began in participants with obesity and other cardiovascular disease risk factors, with topline results expected in the second half of 2025. The company's inflammatory bowel disease (IBD) portfolio includes tamuzimod (formerly VTX002) and VTX958. Tamuzimod is a sphingosine 1 phosphate receptor (S1P1R) modulator being developed for patients with moderately to severely active ulcerative colitis (UC). In the fourth quarter of 2023, Ventyx Biosciences announced positive results from the Phase 2 trial of tamuzimod in patients with moderately to severely active UC.
