Tango Therapeutics, Inc.

TNGX·NASDAQ·Life Sciences·CIK 0001819133

Tango Therapeutics discovers and develops targeted therapies for cancer by focusing on specific genetic changes that drive the disease. The company uses its target and drug discovery platforms to find new disease-relevant targets and create medicines for patient groups with significant unmet medical needs. Its small molecule drugs are designed to selectively kill cancer cells with specific genetic alterations while leaving normal cells unharmed. Tango Therapeutics is currently developing two MTAP-deleted selective PRMT5 inhibitors: vopimetostat (TNG462) and TNG456. Vopimetostat is for non-CNS cancers and is being developed as a single therapy and in combination with RAS inhibitors. TNG456 is a next-generation, brain-penetrant PRMT5 inhibitor for CNS cancers, including glioblastoma (GBM). In October 2025, the company reported positive data from an ongoing Phase 1/2 clinical trial of vopimetostat monotherapy in patients with MTAP-deleted selective cancers. This data showed clinical activity across multiple cancer types with a favorable safety and tolerability profile. Specifically, in second-line MTAP-deleted pancreatic cancer, the data showed a median progression-free survival (mPFS) of 7.2 months and a 25% objective response rate (ORR). This supports the planned start of a second-line pivotal trial in this patient population in 2026. The histology selective cohort, which excludes sarcoma, pancreatic, and lung cancer patients, also showed positive data, with an mPFS of 9.1 months and a 49% ORR. The company is evaluating the development path for this cohort and for selected indications as stand-alone development opportunities. Emerging data from the lung cancer cohort are consistent with expectations, and a safety and efficacy update is anticipated in 2026. The company is also evaluating vopimetostat in combination with RAS inhibitors due to overlapping patient populations, strong preclinical data, and favorable individual drug profiles.
