ImageneBio, Inc. is a clinical-stage biopharmaceutical company focused on developing treatments for patients with immunological, autoimmune, and inflammatory diseases. The company's primary product candidate is IMG-007, a non-depleting anti-OX40 monoclonal antibody. IMG-007 works by specifically binding to the OX40 receptor on activated T cells, which blocks the receptor from binding to the OX40 ligand. IMG-007 is being developed to potentially treat various autoimmune and inflammatory diseases and disorders, with initial evaluation in atopic dermatitis (AD). Key features of IMG-007 include its receptor-targeting mechanism, its non-T cell depleting action (attenuating activated T cell signaling without killing or depleting T cells), and its half-life of approximately 5 weeks. In a Phase 1b/2a clinical proof of concept trial, four-week treatment with IMG-007 showed significant clinical activity sustained for up to 24 weeks. By week 16, 54% of patients achieved EASI-75 (75% reduction in eczema area and severity index), and 31% achieved EASI-90 (90% reduction in eczema area and severity index). The trial also observed durable inhibition of serum inflammatory markers from various T helper cells, including Th1, Th2, and Th17 cells. IMG-007 demonstrated a favorable emerging safety profile and was well-tolerated, with no pyrexia, chills, aphthous or gastrointestinal ulcers, or serious adverse events reported in any clinical studies to date. While IMG-007 is initially being developed for AD, the company believes it has potential for additional indications where OX40 signaling is implicated, such as alopecia areata (AA), asthma, rheumatoid arthritis, and hidradenitis suppurativa.
